Large field CCD system for quantitative imaging of microarrays
JENOPTIK designed and fabricated custom lenses for Dr. Dan Pinkel of the Comprehensive Cancer Center, University of California San Francisco.
The primary design goal of the custom detection optics is to provide an achromatic, well-focused image of the entire object field over a useful wavelength range of 450 to 750 nm.
The set of lenses designed and built for this system include objectives with focal lengths of 75 mm, 105 mm and 150 mm, and a field lens with a focal length of 150 mm.
System format
These lens combinations provide magnifications of 2x, 1.4x and 1x with maximum light collection capabilities of f/2.3, f/3.3 and f/4.7, respectively. The object fields of the objectives are suitable for acquiring images of 12mm, 18mm and 25mm square arrays. The image field has a maximum diameter of 36 mm.
Some of the key aspects to the design are:
- Specimens on 90 μm centers
- 1:10,000 dynamic range
- Broadband fluorescence wavelengths
- Variation ratio < 0.04 across array
- Custom solution driven by need for field flatness and color correction
